View Full Version : adding tessues of mushroom directly to the agar

Hi All ,
I heard that it is possible to directly add mushroom tessue after scraching the cap skin ..

starting from the surface , where exactly to end the extracting operation ? or is it ok to take it from the top of the cap till the bottom (where gills are located) ?

and is there a specific level of agar added to the petri dish ( so the mycelium won't get drown ?

do I have to sink the tessue in the agar medium or just drop it on the surface ? and in what direction (meaning which face of the piece should face to the agar medium when dropping or sinking ??

I too am interested in knowing about this. I've heard the same vagueness. I heard both that this will work fine and that it won't work (I guess it just rots?).

I am also interested in what you do with cultured agar to make it a liquid mycelium. Just add water and suck it through a sterile syringe, or is that damaging to your culture?

I would think you would just need a tiny piece of tissue. This probably sounds ghetto but I take tissue and put directly in LC all the time. I take from the inside of the stem
(where it peels off nicely and is more sterile), dip in peroxide and drop directly into LC (organic barley malt syrup and dextrose mixture)...and it grows like mad...not good for isolation. But you could do this then noc up jar and take a piece of myc from the jar to start sectoring. But then again you could have just taken myc from the where that tissue shroom came from to start with :)

Yeah I really need to know this too. I just bought petri dishes with agar (potato dextrose) and for some reason I am having a problem with collecting spore prints. So I wanted to know how and if I can use shroom tissue to grow mycellium in the petri. Like PF Tek said, I want to grow at an expenential rate!

This is called cloning and you can cut out a piece of flesh from your best looking mushroom, slap it on some agar or into some broth, and it will grow into clones! Best place to take sample is from the base. I personally go for the base of a nice cluster, isolate that on agar, and have huge solid flushes.

Cloning directly to agar is fine and is a good way to keep a good performing strain in storage.

Everytime i added mushroom tissue it just rots. I'm gonna use spores from one of my mushrooms and ill let you know how it does
